罗丹明醛是一种可以与胺,肼或羟胺反应的活性荧光染料。醛基可与pH 5-9的胺,酰肼或羟胺基反应。与胺反应性琥珀酰亚胺基酯基(NHS)不同,醛可以在酸性pH下与N端胺基反应,这是某些生物缀合反应有时需要的条件。醛与胺基反应形成席夫碱。用氢化物进一步还原将形成稳定的C-N键。醛与其他基团之间的反应可实现荧光素染料的位点特异性缀合和标记,使其靶向分子上的所需位置。可以通过普通的荧光仪器在TRITC或Cy3通道下轻松检测缀合罗丹明染料。
