产品介绍

FlASH是一种荧光素衍生物,被修饰为包含两个彼此相距一定距离的砷原子。它由Roger Tsien及其同事在1998年开发。双砷标记技术通过砷与硫醇的高亲和力相互作用起作用。当FlASH结合四半胱氨酸(TC)序列时,其双砷基与Cys-Cys部分快速反应,并且标签变为绿色的高度荧光。双砷标记试剂FIASH是用于标记包含具有Cys-Cys-X1-X2-Cys-Cys氨基酸序列的六氨基酸基序的蛋白质的最小表达标签。最常用的四半胱氨酸是六个氨基酸的Cys-Cys-Pro-Gly-Cys-Cys序列。由于此序列很少出现在内源蛋白中,将序列整合到靶蛋白中会产生一个小的但高度特异性的蛋白标记靶。当与包含四半胱氨酸基序Cys-Cys-Pro-Gly-Cys-Cys的重组蛋白结合时,FIASH会产生强烈的绿色荧光信号。它可以用于检测蛋白质的定位,周转和运输,受体信号传导和内在化。
