产品介绍

Probenecid是位于细胞膜中的有机阴离子转运蛋白的抑制剂。这些转运蛋白通常从细胞中挤出荧光指示剂,从而导致染料保留不良。这种现象通常会导致测定中的高背景,这需要在细胞内良好地保留染料指示剂。使用丙磺舒来抑制转运蛋白的活性,从而减少细胞内染料指示剂的泄漏是降低钙测定的荧光背景的常用方法。每瓶Cat#20060溶于0.3 mL 1M NaOH中,然后稀释到100 mL缓冲液中,使丙磺舒的最终浓度为2.5 mM,用于检测。它可用于防止荧光染料(如Indo-1 AM,Fura-2 AM,Fluo-3 AM,Fluo-4 AM,Fluo-8 AM,Rhod-2 AM和Rhod-4 AM)漏出细胞。我们还提供方便的ReadiUse™水溶性和热稳定丙磺舒,粉末形式(Cat#20061),(Cat#20062)(Cat#20063)。它们使用方便,并且在相同浓度下与游离酸形式一样有效。

产品说明书

操作说明

注意:以下是我们推荐的方案。它仅提供指南,实际应根据您的具体需求进行修改。

1.溶解1瓶Cat#20061在10mL HHBS(1X Hank  – 含20mM Hepes缓冲液,pH7.0)中制备25mM储备溶液。

注意:1 mL 25 mM储备液足以容纳1个平板。 将未使用的储备溶液分装并储存在20℃,避光。 避免反复冻融循环。

2.制备2.5 mM ReadiUse™probenecid缓冲液:将1 ml 25 mM ReadiUse™probenecid原液(来自步骤1)加入9 ml含0.02%~0.04%Pluronic®F-127的HHBS中,并充分混合。

3.为一个细胞板制备染料加载溶液:添加DMSO重组荧光钙染料(如Indo-1 AM,Fura-2 AM,Fluo-3 AM,Fluo-4 AM,Fluo-8 AM,Rhod-2 AM 和Rhod-4 AM)进入2.5 mM ReadiUse™probenecid缓冲液(来自步骤2),并将它们充分混合。 工作溶液在室温下稳定至少2小时。

产品介绍

2’,3′-cGAMP或cGAMP(环鸟苷单磷酸腺苷)是由ATP和GTP的cGAS(cGAMP合酶)在胞浆DNA刺激下在哺乳动物细胞中产生的。cGAS生产的cGAMP含有混合的磷酸二酯键,一个在GMP的2′-OH和AMP的5′-phosphate之间,另一个在AMP的3′-OH和GMP的5′-phosphate之间。由于鸟苷和腺苷之间存在非典型的2′-5’磷酸二酯连接,2’,3′-cGAMP也被称为“非正统”cGAMP。与典型的3’,3′-cGAMP类似,2’3′-cGAMP通过与STING(IFN基因的刺激因子)结合并随后诱导IFN-β的TBK1-IRF3依赖性产生,作为激活先天免疫应答的第二信使。通过对其结构和功能研究表明,非标准2’3′-cGAMP与细菌产生的标准3’3′-cGAMP不同。2’3′-cGAMP以比3’3′-cGAMP拥有更大的亲和力与结合能力。cGAMP还被证明是一种能促进小鼠抗原特异性抗体的产生和T细胞反应的有效试剂。cGAMP具有抗病毒功能,它可以通过被动扩散穿过细胞膜,对邻近细胞产生一些影响。金畔生物是AAT Bioquest的中国代理商,为您提供最优质的2’,3’-cGAMP, 钠盐。
