AF488 由 AAT Bioquest 制造,具有与 Alexa Fluor® 488 相同的化学结构(Alexa Fluor® 是 ThermoFisher 的商标)。 AF488 NHS 酯(琥珀酰亚胺酯)与 Alexa Fluor® 488 NHS 酯(来自 ThermoFisher 的#A20000 和 A20100)具有相同的分子。它是一种亮绿色荧光染料,最适合与 488 nm 氩激光一起使用。 AF488 染料是水溶性的,从 pH 4 到 pH 10 对 pH 不敏感。AF488 NHS 酯(或琥珀酰亚胺酯)是将这种染料与蛋白质或抗体结合的最方便的胺反应形式。尽管 Alexa Fluor® 488 NHS Ester 是最常见的绿色荧光标记染料之一,但其纯度一直颇具挑战性。 ThermoFisher 提供的纯度约为 50-60%,而其他一些供应商提供的纯度甚至更低。在所有杂质中,我们注意到 AF488 的 Bis-NHS 酯是非常有害的,因为它会产生非荧光产物。这种高纯度 AF488 NHS 酯不含有害的 AF488 Bis-NHS 酯杂质,最低纯度为 >75%。
